    目的:分析苹果酸酶3(malic enzyme 3,ME3)在肝细胞肝癌(hepatocellular carcinoma,HCC)组织以及HCC细胞系中的表达水平,并研究ME3对HCC细胞增殖、迁移和侵袭等方面的影响及其可能机制。方法:使用癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库分析 ME3 在 HCC 组织中的表达;使用 RT-qPCR 和 Western blot 技术检测 HCC 细胞系中 ME3 的 mRNA以及蛋白的表达水平;在下调或者上调HCC细胞系中ME3后,借助CCK-8、平板克隆法、Transwell、侵袭实验研究ME3在肝癌细胞增殖、迁移和侵袭方面的作用;运用Western blot检测PI3K/AKT信号通路相关蛋白的表达情况。结果:与癌旁正常组织和正常肝细胞相比,ME3在人类HCC组织和HCC细胞系中表达显著升高,降低ME3表达能减弱HCC细胞的增殖、迁移及侵袭,过表达ME3则会导致相反结果;此外,ME3可以激活PI3K/AKT信号通路。结论:ME3在HCC组织中呈高表达,促进HCC细胞增殖、迁移以及侵袭,并可能通过启动PI3K/AKT信号通路在HCC中发挥其促癌基因的作用。

    Abstract:

    Objective:To analyze the expression of malic enzyme 3(ME3)in hepatocellular carcinoma(HCC)tissues and celllines, and explore the effects of ME3 on the proliferation,migration,and invasion of HCC cells,as well as its underlying mechanism. Methods:The Cancer Genome Atlas(TCGA)database was used to study the expression of ME3 in HCC tissues. RT - qPCR and Western blot were performed to detect the mRNA and protein expression levels of ME3 in HCC cells. After knocking down or over expressing ME3 in HCC cell lines,the effects of ME3 on the proliferation,migration and invasion of HCC cells were detected by CCK-8 assay,colony formation assay,transwell assay,and invasion assay. Western blot was used to detect the expression of proteins related to the PI3K/AKT signaling pathway. Results:Compared with adjacent normal tissues and normal liver celline,ME3 was significantly up-regulated in HCC tissues and cell lines. Knockdown of ME3 markedly inhibited the ability of proliferation,migration and invasion of HCC cells,while overexpression of ME3 resulted in the opposite effects. Furthermore,we found that the activity of PI3K/AKT signaling pathway was significantly activated by ME3. Conclusion:ME3 is highly expressed in HCC tissues,promoting the proliferation, migration and invasion of HCC cells,and may function as an oncogene in HCC by activating the PI3K/AKT signaling pathway.
