    目的:探究羧肽酶E(Carboxypeptidase E,CPE)对高糖培养的小鼠肾小球系膜细胞(Mice mesangial cell,MMC)细胞增殖、周期及凋亡的影响。方法:PCR法扩增CPE基因片段,将其插入表达载体 pcDNA3.1质粒中以构建重组质粒pcDNA3.1-CPE。将质粒转染MMC细胞,通过蛋白质免疫印迹法检测CPE标签蛋白、增殖标志物CyclinD1、PCNA以及凋亡标记物Caspase-3的表达;采用平板克隆、CCK-8法以及流式分析法检测过表达CPE对高糖培养的MMC细胞增殖,周期以及凋亡能力的影响。结果:成功构建CPE过表达的MMC细胞。平板克隆以及CCK-8实验表明,过表达CPE可以抑制MMC细胞的增殖能力。流式细胞学检测表明与对照组相比,CPE过表达明显促进了细胞凋亡;细胞周期中G1期细胞明显增加,S期细胞明显减少,差异有统计学意义(P<0.01)。结论:过表达CPE可以抑制高糖培养的MMC细胞增殖并诱导细胞凋亡。

    Abstract:

    Objective: To investigate the effects of Carboxypeptidase E (CPE) on the cell proliferation, cell cycle and apoptosis of mice mesangial cells (MMC) cultured in high glucose. Methods: The CPE gene fragment was amplified by PCR and inserted into the expression vector pcDNA3.1 plasmid to construct the recombinant plasmid pcDNA3.1-CPE. The plasmid was transfected into MMC, and the expression of CPE-tagged protein, proliferation marker CyclinD1, PCNA and apoptosis markers Caspase-3 were detected by Western blotting; plate cloning ,CCK-8 method and flow cytometry were used to detect the effect of overexpressed CPE on the proliferation, cell cycle and apoptosis of MMC cells cultured in high glucose. Results: pcDNA3.1-CPE over-expression plasmid were successfully transfected into MMC. plate cloning and CCK-8 showed that overexpression of CPE could inhibit the proliferation of MMC. Flow cytometry showed that compared with the control group, CPE over-expression significantly promoted the cell apoptosis, G1 phase cells increased significantly as well as S phase cells decreased obviously, the difference was statistically significant (P<0.01). Conclusion: CPE can inhibit the proliferation of MMC cultured in high glucose and induce the cell apoptosis.
