    目的:探讨东莨菪碱对小鼠边界细胞编码及空间记忆能力的影响。方法:将健康雄性2月龄C57BL/6小鼠随机分为生理盐水对照组和东莨菪碱组,使用morris水迷宫实验分析两组的空间记忆能力,随后向压后皮质(retrosplenial cortex, RSC)植入32通道微丝电极;手术后恢复7天,进行旷场实验并同时记录电信号,通过计算分离出边界细胞。随后通过腹腔给药方式,给与小鼠25 μg/kg 东莨菪碱或等体积的生理盐水,24 h后进行旷场实验并同时记录电信号,与给药前记录到的边界细胞进行对比。结果:生理盐水对照组和东莨菪碱组水迷宫实验的学习曲线有明显差异(P < 0.05),目标象限时间、穿越平台次数均有明显差异(P < 0.05)。注射东莨菪碱或生理盐水24 h后,小鼠旷场总路程无明显差异(P > 0.05);注射东莨菪碱对小鼠边界细胞放电频率无显著性影响(P > 0.05);注射东莨菪碱后,边界细胞系数(Earth Mover’s Distance, EMD)显著增加(P < 0.05),而注射生理盐水无明显影响(P > 0.05)。结论:东莨菪碱可能是通过影响小鼠边界细胞编码从而影响其空间记忆能力的。

    Abstract:

    Objective: To investigate how scopolamine affects border cell coding and spatial memory of mice. Methods: Healthy male 2-month-old C57BL/6 mice were randomly divided into saline control group and scopolamine group. Using morris water maze to evaluate spatial memory ability. Then 32 channel microwire electrodes were implanted into retrosplenial cortex (RSC). After 7 days of recovery, the open field experiment was carried out and the electrical signals were recorded at the same time. Border cell were isolated after analysis. Then 25 μg/kg scopolamine or equivalent volume of normal saline was intraperitoneally injected. After 24 hours, the open field experiment was carried out and the electrical signals were recorded at the same time. The changes of border cells recorded before were analyzed. Results: There were significant differences in the learning curve of water maze experiment between saline control group and scopolamine group (P < 0.05), the time in the target quadrant (P < 0.05) and the number of crossing platform (P < 0.05). After intraperitoneal injection of scopolamine or normal saline for 24 hours, the total distance of mice in open field had no significant difference (P > 0.05); Injection of scopolamine had no significant effect on the firing frequency of border cells in mice (P > 0.05); The earth mover's distance (EMD) was significantly increased after scopolamine injection (P < 0.05), while normal saline injection had no significant effect (P > 0.05). Conclusion: Scopolamine may affect the ability of spatial memory by affecting boundary cell coding in mice.
