Abstract:Objective: To investigate the relationship between nuclear translocation of peroxiredoxin 4 (PRDX4) and cell senescence phenotype in ovarian granulosa cells (GCs) under aging condition. Methods: Female C57BL / 6 mice aged 6 weeks (young group) and 9 months (aged group) were selected respectively. Ovarian tissues were taken for immunofluorescence staining to observe the nuclear translocation of PRDX4 in GCs. At the same time, the human ovarian GCs line KGN was cultured and the in vitro aging model was induced by thapsigargin (Tg). The optimal induction conditions were screened by CCK-8 method. The expression levels of senescence-associated proteins (P16, P21, P53) and senescence-associated secretory phenotype (SASP) factors (IL-1α, IL-1β, IL-6, IL-8) were detected by Western blot (WB) and real-time fluorescence quantitative PCR (RT-qPCR) to verify the aging model. On this basis, the effect of senescent phenotype on PRDX4 nuclear translocation was further verified. Results: Immunofluorescence results showed that, compared with the young group, the fluorescence signal in the nucleus of PRDX4 in ovarian GCs of the aged group was significantly enhanced (Mean difference = 24.14,95 % CI: 18.06-30.22, P<0.01). In vitro, PRDX4 was mainly localized in the cytoplasm of normal KGN cells. After Tg-induced aging, the expression levels of P16, P21, and P53 proteins in the aging group were significantly increased (Fold change ( FC ) = 1.64、1.28、1.97,95 % Ci:1.16 ~ 2.12、1.07 ~ 1.50、1.52 ~ 2.42,P<0.05,P<0.01), and the mRNA expression levels of SASP-related factors were also significantly up-regulated (FC=14.69、1.46、1.30、4.83,95% CI:12.80~16.58、1.14~1.78、1.02~1.57、3.73~5.92,P<0.001,P<0.0001), confirming that the aging model was successfully constructed. Nuclear-cytoplasmic separation experiments further showed that the content of PRDX4 protein in the nucleus of the aging group was significantly higher than that in the young group(FC = 1.59,95% CI: 1.21 ~ 1.97,P<0.01), while the corresponding decrease in the cytoplasm(FC = 0.54,95% CI: 0.35 ~ 0.84,P<0.01)indicated that PRDX4 was transferred from the cytoplasm to the nucleus. Conclusion: Aging can induce the transfer of PRDX4 from cytoplasm to nucleus in ovarian GCs, and the change of nuclear translocation may be involved in the regulation of ovarian aging process.
