目的:探讨哮喘大鼠肺内感觉神经-内分泌细胞分布和密度的变化。方法:20只Wister大鼠随机分成正常组和哮喘组,每组10只。采用免疫组化和计算机图像分析观察大鼠肺内降钙素基因相关肽(calcitonin gene-related peptide,CGRP)和血管活性肠肽(vasoactive intestinal peptide,VIP)免疫反应阳性纤维以及蛋白基因产物9.5(protein gene product 9.5,PGP9.5)和突触素(Synaptophysin,SYN)阳性细胞的变化。结果:①哮喘大鼠肺内CGRP和VIP阳性纤维的分布和密度均发生了显著的变化,CGRP阳性纤维显著增多,染色加深,相对阳性染色面积和灰度值与对照组相比,差异有统计学意义(P < 0.01);VIP阳性纤维明显缺乏,相对阳性染色面积和灰度值与对照组相比,差异有统计学意义(P < 0.05或P < 0.01)。②哮喘大鼠肺内PGP9.5、SYN阳性细胞数量显著增多,染色加深,PGP9.5、SYN阳性细胞率和光密度值与对照组相比,差异有统计学意义(P < 0.01)。结论:哮喘大鼠肺内感觉神经纤维-内分泌细胞的分布和密度存在异常,这可能是导致相应的神经肽含量变化的直接原因,神经内分泌调节失衡,从而促进了哮喘的持续和发展。
Objective:To investigate the changes of distribution and density of sensory nerve and neuroendocrine cells in the lungs of asthmatic rats. Methods:Twenty Wistar rats were divided into asthma group and control group. Immunohistochemistry and computer image analysis were used to observe the changes of the calcitonin gene-related peptide(CGRP) and vasoactive intestinal peptide(VIP) immunoreactive(IR)nerve fibers,protein gene product 9.5(PGP9.5)and Synaptophysin(SYN)positive cells in this study. Results:①There were significant changes in distribution and density of CGRP-IR and VIP-IR nerve fibers in airways of asthma group. The relative positive staining areas and grey levels of CGRP-IR nerve fibers increased significantly in asthma group compared with control group(P < 0.01). But the relative positive staining areas and grey levels of VIP-IR nerve fibers decreased significantly in asthma group compared with control group(P < 0.05 or P < 0.01).②The rate of positive cells and mean optical density(MOD) of PGP9.5 and SYN increased significantly in asthma group compared with control group(P < 0.01). Concolusion:The distribution and density of sensory nerve and neuroendocrine cells were abnormal in the lungs of asthmatic rats,which might immediatly cause the changes of neuropeptides. The adjustment of neuroendocrine was unbalanced,which could promote the persistence and development of asthma.
