    目的:探讨N-甲基-D-天冬氨酸(NMDA)受体拮抗剂MK-801拮抗阿尔茨海默病(AD)果蝇模型中Aβ42诱导的神经毒作用-方法:建立转Aβ42基因果蝇AD模型,用NMDA受体拮抗剂MK-801分别干预正常对照果蝇和转Aβ42基因AD模型果蝇,在整体动物水平上进行果蝇行为学检测,分别检测各组果蝇的嗅觉短期记忆能力-攀爬能力和平均寿命-结果:与Aβ42转基因组果蝇相比,Aβ42转基因果蝇喂食MK-801组的学习记忆能力-攀爬能力-平均寿命均有显著改善(P < 0.05),正常对照组和正常对照+MK801组的行为学检测无统计学差异-结论:在转Aβ42基因AD果蝇模型中,NMDA受体拮抗剂MK-801的干预有明显的神经保护作用,能减轻Aβ42诱导的神经毒作用-

    Abstract:

    Objective:To explore the neuroprotective effects of the N-methyl-D-aspartate(NMDA) receptor antagonist MK-801 on Aβ42 transgenic drosophila model of Alzheimer’s disease. Methods:Drosophila was designed into four groups:normal control,normal drosophila exposed to MK-801,Aβ42 transgenic drosophila and Aβ42 transgenic drosophila exposed to MK-801. And the olfactory associative learning assay,climbing assay and survival assay were performed in four groups of drosophila. Results:Compared with Aβ42 transgenic drosophila group,the associative learning ability,climbing ability and life span of Aβ42 transgenic drosophila applied with MK-801 group were significantly improved. Conclusion:In Aβ42 transgenic drosophila model of Alzheimer’s disease,MK-801 can reduce the neurotoxic effect induced by Aβ42.
