    目的:研究经Ⅰ型胶原-壳聚糖改性的PLGA膜的生物相容性及力学特性,研制出新型人工硬脊膜替代材料-方法:制作PLGA膜(膜Ⅰ)-PLGA/Ⅰ型胶原/复合膜(膜Ⅱ)-PLGA/Ⅰ型胶原/壳聚糖(9 ∶ 1)复合膜(膜Ⅲ)-PLGA/Ⅰ型胶原/壳聚糖(5 ∶ 5)复合膜(膜Ⅳ),并行接触角-吸水率-细胞毒性实验及材料力学实验的研究-结果:接触角:膜Ⅱ < 膜Ⅲ < 膜Ⅳ < 膜Ⅰ,P < 0.01;吸水率:膜Ⅰ < 膜Ⅳ < 膜Ⅲ < 膜Ⅱ,P < 0.01;细胞毒性实验示:第1天,各膜组间OD值差异无统计学意义,P > 0.05-第3-7天,膜Ⅰ与Ⅱ-Ⅲ各组间,膜Ⅲ与膜Ⅳ组间差异有统计学意义,P < 0.05-PLGA膜经Ⅰ型胶原和壳聚糖改性后,可以促进细胞在膜上的黏附-贴壁能力-各膜的力学性能比较,膜Ⅰ与膜Ⅱ-Ⅲ-Ⅳ之间差异有统计学意义,P < 0.05,膜Ⅱ-Ⅲ和Ⅳ组间的差异无统计学意义-结论:PLGA膜的表面复合Ⅰ型胶原/壳聚糖(9 ∶ 1)可以提高复合膜的生物相容性和力学特性-

    Abstract:

    Objective:To study the biocompatibility and mechanical properties of PLGA membrane modified by Type-Ⅰcollagen and Chitosan,and to develop a novel tissue engineering materials as artificial spinal dura mater. Methods:PLGA membrane(membraneⅠ),PLGA/ Type-Ⅰcollagen composite membrane(membraneⅡ),PLGA/ Type-Ⅰcollagen/ Chitosan(9 ∶ 1) composite membrane(membrane Ⅲ) and PLGA/ Type-Ⅰcollagen/ Chitosan(5 ∶ 5) composite(membrane Ⅳ) were produced through a certain process. Contact angle,absorption rate,cytotoxicity study and determination of mechanical properties were used to research all type membranes. Results:Contact angle:membraneⅡ < membraneⅢ < membraneⅣ < membraneⅠ,P < 0.01; Absorption rate:membraneⅠ < membrane Ⅳ < membrane Ⅲ < membrane Ⅱ,P < 0.01;Cytotoxic experiment(MTT method):at the 1st day,the OD value between each group didn’t have significant difference,P > 0.05. At the 3rd and the 7th day,there was significant difference between membrane Ⅰand Ⅱor Ⅲ,membrane Ⅲ and Ⅳ,P < 0.05. After being modified by Type-Ⅰcollagen and Chitosan, PLGA membrane striking enhanced the adhesion and proliferation of L929 cell,and the comparison between all type membranes showed that there was significant difference between membrane Ⅰand other type membranes,P < 0.05,and there was no significant difference between membrane Ⅱ,Ⅲ and Ⅳ. Conclusion:With type-Ⅰcollagen and Chitosan(9 ∶ 1) appending to the exterior, PLGA can enhance the biocompatibility and Mechanical properties of membrane.
