    目的:观察leptin对人成熟脂肪细胞中NYGGF4基因表达的调节作用-方法:体外培养人前体脂肪细胞,在诱导人前体脂肪细胞分化成熟的基础上,应用100 ng/ml人重组leptin分别干预成熟脂肪细胞6-24 h,采用实时荧光定量RT-PCR技术检测成熟脂肪细胞中NYGGF4基因mRNA的表达水平-结果:100 ng/ml leptin干预人成熟脂肪细胞6 h后,NYGGF4基因mRNA的相对表达量为0.000 476 ± 0.000 178,显著低于对照组0.001 14 ± 0.000 275,P < 0.05;干预24 h NYGGF4基因mRNA的相对表达量为0.000 835 ± 0.000 211,也显著低于对照组0.001 419 ± 0.000 193,P < 0.05-结论:leptin能显著下调成熟脂肪细胞中NYGGF4基因的表达-

    Abstract:

    Objective:To explore the regulative role of leptin on NYGGF4 gene expression in matured human adipocytes. Methods:Human preadipocytes were cultured in vitro and differentiated into matured adipocytes. The matured adipocytes were treated with 100 ng/ml leptin for 6 hours and 24 hours,respectively. Total RNA from these adipocytes was extracted and the levels of NYGGF4 mRNA expression were evaluated by real-time quantitative RT-PCR. Results:Treatment with 100 ng/ml leptin for 6 hours or 24 hours significantly decreased NYGGF4 gene mRNA expression in human matured adipocytes (P < 0.05). Conclusion:Leptin could downregulate the expression of NYGGF4 gene mRNA in human matured adipocytes.
