Abstract:Objective:To investigate the feasibility of microbubble mediating Ang-1 gene therapy of lower limb ischemia by using ultrasound. Methods:Fifteen rabbits with the left femoral artery ligation were randomly and averagely devidied into 3 groups:group A was locally intramuscular injected with the mixture of plasmid pEGFP/Ang-1 and microbubble and accepted ultrasound local radiation,group B was locally intramuscular injected with the plasmid pEGFP/Ang-1,and group C was injected with NS. Four weeks post-treatment,angiography was applied to evaluate the collateral circulation,and the expression of Ang-1 was evaluate by immunochemical staining. Results:In the matter of the collateral circulation,the degree of brown in cytoplasm,new vascular of the treatment region,group A was superior to the group B,the group B was superior to the group C. Conclusion:Microbubble mediating Ang-1 gene therapy by using ultrasound could promote the collateral circulation and new vessels,it may be a new methods for the treatment of limb ischemia diseases.
