Abstract:Objective:To investigate the effects of Fe2+on the proliferation of the HUVSMC and its secretion of MMP-2 under the condition of normal or high glucose. Methods:Primarily cultured HUVSMC(Passage 3~5)were cultured with the normal and high glucose DEME,and then subjected to control or Fe2+ incubation at various concentrations(0.006 25 g/L,0.012 5 g/L,0.025 g/L,0.05 g/L,0.1 g/L). After 24 h of intervention,HUVSMC proliferation was assessed with MTT method,and MMP-2 secretion was measured by ELISA. Results:Compared to the control group:in normal glucose group,HUVSMC proliferation in 0.012 5~0.1 g/L Fe2+ treatment group was increased(P < 0.01),and the secretion of MMP-2 was promoted only in 0.1 g/L Fe2+ treatment group(P < 0.01);in high glucose group:HUVSMC proliferation in 0.012 5~0.1 g/L Fe2+ treatment group was increased(P < 0.05),and the secretion of MMP-2 was promoted in all experimental groups(P < 0.05);and the proliferation of HUVSMC and level of MMP-2 in high glucose group were higher than those in the normal glucose group(P < 0.05). Conclusion:Fe2+ could stimulate the proliferation of VSMC,as well as the secretion of MMP-2. And high glucose may contribute to that accelerated effects.
