    目的:研究EGF和TGF-α调控舌鳞癌细胞周期分布的信号通路-方法:不同浓度EGF和TGF-α作用体外培养的舌鳞癌细胞株TCA-8113,流式细胞仪检测细胞周期分布,细胞免疫组化法研究p38-MAP kinase-c-myc-c-fos和NF-κB在舌鳞癌细胞中的表达水平-结果:EGF和TGF-α效应类似,与对照组相比,G1/G0期细胞比例下降,S期细胞比例增加,p38-MAP kinase-c-myc和c-fos表达水平下调,NF-κB p65表达水平上调,但量效关系不同-EGF和TGF-α分别上调和下调NF-κB p50表达-结论:EGF和TGF-α对舌鳞癌细胞周期分布的影响差异与NF-κB相关,NF-κB可能与舌鳞癌发生关系密切-

    Abstract:

    Objective:To investigate the signal pathway of EGF and TGF-α modulating the distribution of cell cycle in tongue squamous cell. Methods:Flow cytometric analysis and immunohistochemistry were employed to investigate the cell cycle distribution,and expressed levels of p38-MAP kinase,c-myc,c-fos and NF-κB respectively. Results:EGF and TGF-α showed similar effect on tongue squamous cell,but the dose-effect relationships were different. In comparison with the control,the proportion of cells in G1/G0 declined and that in S phase ascended. The levels of p38-MAP kinase,c-myc and c-fos were down-regulated and NF-κB p65 up-regulated. EGF and TGF-α up- and down-regulated the expressed levels of NF-κB p50 respectively. Conclusion:The distinct expression of NF-κB may be one of the reasons that results in the difference of EGF and TGF-α on the cell cycle distribution. NF-κB is closely related to the occurrence of tongue squamous carcinoma.
