    目的:研究人单核/巨噬细胞系THP-1与人原代脂肪细胞共培养体系中白细胞介素(IL-6)分泌水平的变化,以探讨炎症和肥胖的相关性-方法:建立THP-1与人原代脂肪细胞共培养体系,并用JNK抑制剂干预共培养体系24 h后,用酶联免疫吸附法(ELISA)分别测定单核细胞组-脂肪细胞组-共培养组及共培养加JNK抑制剂组上清中IL-6的水平-结果:共培养组较单核细胞组-脂肪细胞组IL-6水平升高(P < 0.05),共培养加入JNK抑制剂后,IL-6水平有所下降(P = 0.114)-结论:人单核/巨噬细胞系THP-1与人原代脂肪细胞共培养促进炎症因子IL-6的释放,选择性阻断炎症介导的信号传导途径,IL-6的释放减少,对症使用抗炎介质及JNK抑制剂有望成为肥胖治疗的新靶点-

    Abstract:

    Objective:To examine the changes of IL-6 level in the condition of cultured THP-1 with or without human adipocytes,and to explore the relationship between inflammation and obesity. Methods:Co-culture system for monocytic cell line THP-1 and human adipocytes was established through transwell combined with treatment of JNK inhibitor-SP600125. Meanwhile,cultured THP-1 and human adipocytes respectively acted as controls. The accumulation of IL-6 in cultured media supernatant was assayed by enzyme-linked immunosorbant assays (ELISA) for every group. Results:The results of ELLSA showed that the IL-6 was markedly increased in the co-culture of THP-1 and human adipocytes,and the level of IL-6 was decreased when treated with JNK inhibitor. Conclusion:There might be a paracrine loop between the monocytic cell line THP-1 and adipocytes. The level of IL-6 is increased under co-culture condition,while JNK inhibitor could decrease the IL-6 release,which might become a new therapy target for obesity.
