    目的:建立一种简单-安全-高成功率的大鼠急性肾脏缺血再灌注损伤动物模型-方法:应用微型动脉夹夹闭Wistar大鼠双侧肾动脉50 min后松开,于伤后第3-5-7天取肾,经一系列处理后进行HE染色及免疫组化染色,以观察在肾脏近曲小管上皮细胞中是否出现肾损伤分子-1(kidney injury molecule-1,Kim-1)抗原及该细胞形态学改变-结果:该法造模成功率达93.3%,实验组肾脏近曲小管上皮细胞Kim-1抗原表达率上升,同时该细胞有明显细胞形态学改变(P > 0.05)-结论: 应用微型动脉夹夹闭Wistar大鼠双侧肾动脉可制备急性肾脏缺血再灌注损伤模型,且成功率高-方法简单-

    Abstract:

    Objective:To construct a simple,safe,high successful rate model of renal acute ischemia-reperfusion injury in Wistar rats. Methods:Renal artery of Wistar rats was bilaterally clamped with a micro-artery clamp for 50 minutes to make the animal model of renal acute ischemia-reperfusion injury. Then these kidneys were removed at the 3rd,5th,7th day after injury. Immunohistochemistry staining was used to show the distribution and expression of Kim-1 antigen,and the changes of cytomorphology were observed by HE staining. Results:The successful rate of model construction reached to 85% in the experiment. The expression of Kim-1 antigen increaced. Meanwhile,changes of cytomorphology took place. Conclusion:Stable model of renal acute ischemia-reperfusion injury of Wistar rats can be constructed by applying micro-artery clamp to incarcerate bilateral renal arteries,and its successful rate is very high.
