    目的:探讨不同方法建立糖尿病肾病(diabetic nephropathy,DN)模型的差异,以确定建立糖尿病肾病大鼠模型的最佳方案-方法:清洁级健康雌性SD大鼠30只,随机分为3组,每组10只-A组大鼠单侧肾脏切除后腹腔注射链脲佐菌素(streptozotocin,STZ)55 mg/kg,B组大鼠单侧肾脏结扎后腹腔注射STZ 55 mg/kg,C组为假手术对照组,注射等量的缓冲液-大鼠造模后从第1周开始每2周称体重,取血测血糖(Glu)-血肌酐(Scr)-总胆固醇(Chol)和甘油三酯(TG),收集尿测24 h尿蛋白(24Upro)-尿肌酐(Ucr),并计算内生肌酐清除率(Ccr)-11周后,处死大鼠,取肾脏称重并进行肾脏病理-结果:A-B 2组的成模周期相当,B组的存活率高于A组-结论:单侧肾脏结扎后再腹腔注射55 mg/kg STZ可以用来作为建立糖尿病肾病大鼠模型的理想方案-

    Abstract:

    Objective:To investigate the different model construction methods in diabetic nephropathy(DN) SD rats induced by streptozotocin(STZ) and establish the optimal plan. Methods:Thirty SD rats were randomly divided into 3 groups with 10 rats in each group. A group received intraperitoneal STZ after unilateral nephritic excision;B group received intraperitoneal STZ after unilateral nephritic ligation;C group(control group) received buffer solution only. The changes of body weight(BW),glucose(Glu),serum creatinine (Scr),cholesterol(Chol),triglyceride (TG)and 24-urine protein(24Upro),urine crea(Ucr)were detected every 2 weeks and creatinine clearance rate(Ccr) was calculated. After 11 weeks,the rats were sacrificed,and the kidneys were weighed and taken for pathological examination. Results:There was no significant difference in the model construction periods between the groups A and B. The survival rate of the group B is higher than that of the group A. Conclusion:STZ(55 mg/kg) intraperitoneal injection after unilateral nephritic ligation is the optimal method for inducing rat diabetic nephropathy.
