    目的:观察强脉冲光(intense pulsed light,IPL)对小鼠皮肤的胶原纤维及细胞增殖核抗原(proliferating cell nuclear antigen,PCNA)的影响,探讨强脉冲光嫩肤的分子生物学机制-方法:采用能量密度为32 J/cm2强脉冲光照射小鼠背部右侧皮肤,左侧为对照,分别于照射后1-7-14-28天切取皮肤样本,行HE染色,苦味酸-酸性品红染色(VG 染色) 观察皮肤组织及胶原变化,免疫组化检测皮肤PCNA的表达-结果:与对照部位比较,IPL照射对真皮厚度无明显改变(P < 0.05);照射后1-7天两侧皮肤胶原含量无明显改变(P > 0.05),14及28天后照射部位皮肤胶原明显增加 (P < 0.05);PCNA表达于照射后1-7天无明显改变(P > 0.05),14及28天显著增强(P < 0.05)-结论:强脉冲光可促进皮肤胶原含量增加,并可诱导PCNA表达增强-

    Abstract:

    Objective:To observe the effect of intense pulsed light (IPL) on collagens and PCNA expressions in mouse skin and elucidate the biological mechanisms of photorejuvenation. Methods:Mice were irradiated on their right sides of the back with IPL(32 J/cm2),and their left sides were selected as control areas. At day 1,7,14 and 28 after treatment,skin specimens were taken at treated and non-treated areas,respectively. Histological and collagens changes were observed by hematoxylin-eosin-staining and Van Gieson staining. In addition,the expression of PCNA was investigated by immunohistochemistry at day 28 after irradiation. Results:Compared with non-treated regions,dermal thickness of treated regions had no significant change at day 1,7,14 and 28(P > 0.05). Collagens staining were stronger in treated regions than in non-treated areas at day 14 and 28(P < 0.05). The expression of PCNA also increased at day 14 and 28 after irradiation in treated areas(P < 0.05). Conclusion:IPL can enhance the content of collagens and also can increase the expression of PCNA in mouse skin.
