Abstract:Objective:To investigate the expression and significance of C/EBP homologous protein/growth-arrest and DNA damage inducible gene 153(CHOP/GADD153) in preeclampsia. Methods: Semiquantitative RT-PCR,immunohistochemistry and Western-blot were used to investigate the expression of CHOP/GADD153 mRNA and protein levels in placenta of 30 women with preeclampsia and 35 normal pregnant women. Results: ①The expression of CHOP/GADD153 mRNA was significantly higher in preeclampsia group(13.416 ± 1.516) than in the normal pregnant group(2.615 ± 0.659,P < 0.01). ②The mean density of CHOP/GADD153 in preeclampsia placenta(0.156 ± 0.008) was significantly higher than that in the th normal pregnant women group(0.143 ± 0.009,P < 0.01). ③The expression of CHOP/GADD153 protein levels was significantly higher in preeclampsia group(16.210 ± 1.217)than in the normal pregnant group(2.710 ± 0.799,P < 0.01). Conclusion:Up regulation CHOP/GADD153 mRNA and excess expression of CHOP/GADD153 protein levels in placenta may be involved in the pathophysiological processes of preeclampsia.
