    目的:确定骨髓间充质干细胞(bmscs)在异种胸腺内的演化过程,为进一步进行bmscs胸腺修饰诱导异种免疫耐受的研究提供可行性依据-方法:取豚鼠bmscs经分离纯化体外扩增至第3代,并行胎牛血清-fe2o3和4,6-二咪基-4-联苯基吲哚(dapi)标记,标记后的豚鼠bmscs行胸腺注射,7天后进行切片普鲁士蓝-中性红染色和荧光显微镜观察豚鼠bmscs演变过程-结果:bmscs移植至异种胸腺中能够生存,并能转化为多种组织细胞成分,即仍具有多向分化能力-结论:异种bmscs胸腺修饰在诱导异种受体移植免疫耐受方面将是可行的-

    Abstract:

    objective:to observe the proliferation and differentiation of xenogeneic bone mesenchymal stem cells (bmscs) in the thymus and to explore the feasibility of intrathymic inoculation with xenograft-bmscs in inducing immunotolerance. methods:bmscs of guinea pigs were cultured in medium to p3,and labeled with fetal bovine serum-fe2o3 and 4,6-diamidino-2- phenylindole(dapi).the marked bmscs were inoculated into thymuses gland of rats. the thymuses were sliced up and dyed with perls solution seven days later and observed directly under the fluorescenc microscope. results:bmscs survived long term in the micro-environment of the xenogeneic thymuses,and had the ablitity to differentiate into various cell types. the xenogeneic bmscs had the capablilty of multiple rounds of division. conclusion:it is feasible to induce immune tolerance by intrathymic inoculation with xenograft-bmscs.
