目的:探讨miRNA-26a在支气管哮喘患儿及支气管哮喘小鼠模型中的表达-方法:应用real-time PCR法检测支气管哮喘患儿外周血淋巴细胞中miRNA-26a与正常儿童的表达差异-采用卵清蛋白(ovalbumin,OVA)诱导支气管哮喘小鼠模型,通过检测肺泡灌洗液(BALF)中细胞数及组织病理验证模型建立-应用real-time PCR法检测支气管哮喘小鼠模型肺组织miRNA-26a与对照组的表达差异-采用Western blot 法检测小鼠肺组织GSK-3β的表达差异-结果:支气管哮喘患儿外周血淋巴细胞miRNA-26a显著高于正常对照组-OVA诱导的支气管哮喘小鼠模型组BALF中细胞总数及嗜酸性粒细胞数显著高于正常对照组-组织病理显示支气管哮喘模型组炎症细胞浸润明显高于正常对照组-支气管哮喘小鼠模型组肺组织miRNA-26a显著高于正常对照,GSK-3β蛋白水平明显低于正常对照组-结论:miRNA-26a在支气管哮喘患儿及支气管哮喘小鼠模型中高表达-
Abstract:
Objective: To investigate the differencial expression of miRNA-26a in allergic asthma children and murine asthma model. Methods: Expressions of miRNA-26a in allergic asthma children and ovalbumin-induced mouse asthma model were analyzed by real-time PCR. The number of cells in the bronchoalveolar lavage fluid (BALF) of murine asthma model was acessed. The expression of GSK-3β in murine asthma model was acessed by Western blot. Results: miRNA-26a was up-regulated in allergic asthma children and murine asthma model compared with normal controls. The total cell number and acidocyte number in BALF from murine asthma model were significant higher than control. Enhanced eosinophilic inflammation in lung was observed in murine asthma model. Protein level of GSK-3β in murine asthma model was lower than control. Conculsion: miRNA-26a expression level was higher in allergic asthma children and murine asthma model than controls.
