目的：验证程序性细胞死亡因子4 (programmed cell death 4，PDCD4)在小鼠雄性生殖细胞中的表达。方法：RT-PCR检测PDCD4在不同周龄小鼠睾丸的基因表达情况；Western blot法检测PDCD4在不同周龄小鼠睾丸中蛋白表达水平；免疫荧光法检测PDCD4在雄性生殖嵴以及在不同周龄小鼠睾丸的定位情况。结果：RT-PCR和Western blot结果均显示PDCD4在0周龄小鼠睾丸中开始有表达，并持续表达至成年；免疫荧光显示PDCD4在胚胎期雄性小鼠生殖细胞中也有表达；在出生后0周和1周龄小鼠睾丸中，PDCD4特异表达于精原细胞；在2周龄小鼠睾丸中，PDCD4分布于精原细胞和精母细胞；而在3、4和5周龄小鼠睾丸组织中，PDCD4主要分布于精原细胞、精母细胞与圆形精子细胞。结论：PDCD4分布于胚胎期和出生后各阶段生精细胞，这为进一步研究PDCD4在精子发生中的作用打下基础。
Abstract:
Objective：To validate PDCD4 expression in male mouse germ cells. Methods：We verified Pdcd4 expression in testes of different age mice via RT-PCR and Western blot. The localization of PDCD4 in male reproductive ridge and testis in different age groups was detected by immunofluorescence. Results：RT-PCR and Westen bolt results showed that PDCD4 was expressed in the 0-week-old mouse testes and continued to express till adulthood； Immunofluorescence showed that PDCD4 was expressed in embryonic male germ cells. PDCD4 was specifically expressed in spermatogonia in the testis of 0 week and 1 week old mice after birth；In the 2-week-old mouse testes，PDCD4 was distributed in spermatogonia and spermatocytes；In the 3 weeks，4 weeks and 5 weeks old mice testicular tissue，PDCD4 mainly distributed in spermatogonia，spermatocytes and round spermatids. Conclusion：PDCD4 is distributed in each level of male germ cells，which lay the foundation for further study of the role of PDCD4 in spermatogenesis.
