    摘要:

    目的:探讨高糖对人肾小球系膜细胞细胞间黏附分子1(ICAM-1) 表达的影响及茶多酚对其干预作用。方法:将培养的系膜细胞分为正常对照组、高糖组、茶多酚干预组和茶多酚对照组,分别在0、12、36 h采用RT-PCR 法和免疫细胞化学法检测茶多酚对高糖条件下系膜细胞内ICAM-1 mRNA及蛋白表达的影响。结果:高糖组系膜细胞ICAM-1 mRNA和蛋白的表达比正常对照组均增加(P < 0.05);茶多酚干预组比高糖组有明显下降(P < 0.05)。结论:高糖可导致人肾小球系膜细胞ICAM-1表达的增加,而茶多酚可有效干预此效应。

    Abstract:

    Objective:To investigate the effects of high glucose on the expression of ICAM-1mRNA in human mesangial cells and the intervention of tea polyphenols(TPs). Methods:Human mesangial cells were cultured in DMEM media containing high glucose with or without TPs for 0,12,36 hours,respectively. The mRNA and protein expression of ICAM-1 were measured by RT-PCR and immunocytochemistry. Results:The mRNA and protein expression of ICAM-1 of the cells increased significantly in high glucose medium(P < 0.05). However,TPs inhibited the expression(P < 0.05). Conclusion:High glucose contributes to the increase of ICAM-1 expression in human mesangial cells. TPs can reduce high glucose-induced expression of ICAM-1.
