    摘要:

    目的:用高效液相(HPLC)法测定兔眼房水中醋甲唑胺浓度,提供其药动学依据。方法:以10%三氯醋酸沉淀兔眼房水中的蛋白质,采用μ1BondapakTM C18柱(3.9 mm × 300 mm,10 -滋m),流动相为甲醇 ∶ 水(25 ∶ 75,V/V),检测波长为290 nm,流速1 ml/min,柱温40℃。结果:兔眼房水中醋甲唑胺浓度在0.05~10.00 -滋g/ml范围内呈良好的线性关系,r = 0.9982,方法的相对回收率101.7%,绝对回收率82.7%,日内和日间RSD小于10%。结论:高效液相法灵敏度高,操作简单快速,结果可靠,适合于眼房水醋甲唑胺药动学研究。

    Abstract:

    Objective:To establish a practical HPLC method to determine the concentration of methazolamide in aqueous humor of rabbit eyes for the research of methazolamide drug delivery system. Method:Protein was deposited by 10%(V/V) trichloroacetic acid. Methazolamide was extracted from aqueous humor by using C18 column and mobile phase of methanol ∶ water(25 ∶ 75,V/V). The column temperature was maintained at 40℃, methazolamide was detected at 290 nm, and the flow rate was 1.0 ml/min. Results:A good linear relationship was obtained between the concentration of methazolamide from 0.05 -滋g/ml to 10 -滋g/ml, r = 0.9982. The Relative recovery and absolute recovery of methazolamide were 101.7% and 82.7%, respectively; and RSD of intra-day and inter-day were less than 10%. Conclusion:The improved method could significantly enhance the detective sensitivity without damage to the column and increase the accuracy. The procedure was simple, sensitive, and suitable for determination of methazolamide in aqueous humor with good practicability.
