      目的:探讨ERK通路在血小板源性生长因子-BB(PDGF-BB)诱导的大鼠肾小球系膜细胞细胞外基质(ECM)合成中的作用?方法:将体外培养大鼠肾小球系膜细胞(HBZY-1)分为7组:对照组;PDGF-BB刺激组(终浓度分别为10?25和50 ng/ml);PDGF-BB和ERK通路抑制剂UO126共刺激组(PDGF-BB 25 ng/ml+UO126 5 ?滋mol/ml,PDGF-BB 25 ng/ml+UO126 10 ?滋mol/ml);UO126 10 ?滋mol/ml刺激组?应用逆转录-聚合酶链反应(RT-PCR)法检测肾小球系膜细胞基质金属蛋白酶-2(MMP-2)?纤溶酶原激活物抑制物-1(PAI-1)和纤连蛋白(FN) mRNA的相对表达量?结果: PDGF-BB能剂量依赖性的上调 PAI-1 和FN mRNA的表达(P < 0.05),同时下调 MMP-2 mRNA的表达(P < 0.05)?U0126则呈剂量依赖性地抑制PDGF-BB诱导的肾小球系膜细胞PAI-1 和FN mRNA的表达(P < 0.05),促进MMP-2 mRNA的表达(P < 0.05)?结论:PDGF-BB可能通过ERK通路作用于大鼠肾小球系膜细胞,抑制细胞外基质的降解,促进其FN的合成,从而参与肾小球硬化的过程?

      Objective:To explore the role of ERK signal pathway in the extracellular matrix(ECM) synthesis of rat mesangial cells treated with PDGF-BB in vitro. Methods:Cultured rat mesangial cells were divided into seven groups:control cells; treated with PDGF-BB(10,25,50 ng/ml,respectively);treated with PDGF-BB(25 ng/ml) and UO126(5,10 ?滋mol/ml,respectively);treated with UO126 10 ?滋mol/ml. The expression of fibronectin(FN),matrix metalloproteinases-2(MMP-2) and plasminogen activator inhibitor-1(PAI-1) mRNAs were detected by RT-PCR. Results:In mesangial cells,PDGF-BB downregulated MMP-2 mRNA expression(P < 0.05),upregulated PAI-1and FN mRNAs expression in a dose-dependent way(P < 0.05). The changes of FN,MMP-2 and PAI-1 mRNA induced by PDGF-BB can be inhibited by UO126 in a dose-dependent fashion(P < 0.05). Conclusion:PDGF-BB can inhibit the degradation of extracellular matrix and stimulate the synthesis of FN in rat mesangial cells through ERK siginal pathway.
