    目的:建立紫外损伤的细胞模型并探讨中药杜仲抗紫外线损伤的效果-方法:DMEM液培养的胚鼠细胞经紫外线照射处理建立细胞损伤模型,比较不同浓度杜仲提取液预处理后细胞存活率;比较对照组-UC组(紫外照射30 min)-UE组(杜仲培养并紫外照射30 min)细胞形态学的不同,用噻唑蓝(MTT)法测细胞存活率,用流式细胞仪测定细胞线粒体跨膜电位,用化学发光法测定细胞线粒体ATP含量-结果:紫外照射30 min是建立细胞损伤模型的最佳时间,杜仲浓度在0.4 mg/ml时能够最大程度地保护细胞;电镜观察发现紫外照射30 min后细胞发生凋亡;线粒体跨膜电位检测结果显示UE组比UC组下降的幅度要小;ATP含量由高至低排列依次为:UC组,UE组和对照组-结论:成功构建了紫外损伤细胞模型,紫外照射30 min为适宜时间;0.4 mg/ml杜仲提取液预处理能够有效地抗紫外损伤-

    Abstract:

    Objective:To establish a cell model for ultraviolet damage to the skin and to study whether Eucommia has a protective effect against the UV-induced damage. Methods:Through the method of MTT,the most suitable Ultra Violet Radiation(UVR) time was screened out to best establish the model and found out the optimal concentration of Eucommia Ulmoides Oliv. Three groups were set in this experiments,control,UC and UE. UC were treated by a 30-minute-UVR,UE were treated by Extracts from Eucommia Ulmoides Oliv culture and then a 30-minute-UVR. Microscope and electromicroscope were used to morphologically observe the extent of damage by UVR. Further assessment was executed on the membrane potential of mitochondria and the content of ATP to better understand the extent of mitochondria damage. Results:30-minute-UVR was an ideal choice for the model. Extracts from Eucommia Ulmoides Oliv at the concentration of 0.4 mg/ml was demonstrated the most effective. Electromicroscrope showed cell would decline after a 30-minute-UVR. The protective effect on cell in UE group could be observed morphologically. The result of membrane potential of mitochondria was that UE declined by less extent than UC group compared with the control group.The ATP content ranging from high to low was UC,UE and control group. Conclusion:A damaged cellular model was established by 30 minutes of UV radiation,and the results showed the photoprotective function of eucommia at the concentration of 0.4 mg/ml.
