    摘要:

    目的:研究过氧化物酶体增殖物激活受体γ(PPAR-γ)激动剂对瘦素刺激的血管平滑肌细胞(VSMCs)增殖效应的影响。方法:采用原代细胞培养方法建立大鼠VSMCs 细胞模型;以四甲基偶氮唑盐(MTT)比色法观察不同浓度的瘦素及PPAR-γ激动剂吡格列酮对VSMCs 增殖的影响。结果:瘦素能显著促进VSMCs 增殖,促增殖效应在72 h 末达峰值且瘦素浓度在0~100 ng/ml之间呈剂量依赖关系,差异有统计学意义(P < 0.05);而PPAR-γ激动剂吡格列酮能抑制瘦素诱导的VSMCs 的增殖效应,最佳浓度在100 μmol/L时(P < 0.01)。结论:瘦素能刺激VSMCs增殖,而PPAR-γ激动剂能够抑制瘦素的促增殖作用,有望用于干预动脉粥样硬化的发生发展,上述实验结果可能与纠正瘦素抵抗的机制有关

    Abstract:

    Objective:To observe the effect of peroxisome proliferator-activated receptor γ agonist(pioglitazone) on leptin-induced proliferation of vascular smooth muscle cells(VSMCs). Methods:By using primary cell culture and MTT colorimetry,the effects of leptin and peroxisome proliferator-activated receptor γ agonist were detected at various concentrations on VSMCs proliferation. Results:Leptin can significantly promote VSMCs proliferation in a dose-and time-dependent manner,whose maximum effect appears at 100 ng/ml and 72 hours(P < 0.05). In additiom,pioglitazone suppressed leptin-induced VSMCs proliferation,especially at 100 μmol/L.(P < 0.05). Conclusion:Leptin can induce the proliferation of VSMCs,and pioglitazone can inhibit this effect. Pioglitazone might be used to treat atherosclerosis, and the mechanism should be involved in that pioglitazone could relieve the leptin resistance.
