      目的:探讨苦参碱?γ干扰素(IFN-γ)对博莱霉素诱导的大鼠肺纤维化的干预作用及其可能的机制?方法:SD雄性大鼠60只,分为健康对照组?肺纤维化模型组?苦参碱干预组?IFN-γ干预组,各组分别于7 d?14 d?28 d处死5只,行HE染色及Masson染色半定量判断肺组织肺泡炎及肺纤维化程度?免疫组化测肺组织TGF-β1?CTGF?I型胶原及Ⅲ型胶原的蛋白水平表达?结果:苦参碱干预组?IFN-γ干预组大鼠肺泡炎?肺纤维化程度均较模型组好转?与模型组相比,苦参碱?IFN-γ都能够抑制肺组织中的TGF-β1?CTGF?I型胶原及Ⅲ型胶原的表达(P < 0.05)?结论:苦参碱及IFN-γ对大鼠肺纤维化模型均有一定的疗效,其作用机制可能是通过抑制TGF-β1和CTGF的表达来减少Ⅰ型胶原及Ⅲ型胶原的生成?

      Objective:To evaluate the therapeutic effect and mechanism of matrine and IFN-γin Bleomycin-induced pulmonary fibrosis of rats. Methods:Male SD rats(n = 60) were randomly divided into four groups:(A)normal group,(B)Bleomycin-induced pulmonary fibrosis model group,(C)Matrine group,(D)IFN-γ group.On days 7,14 and 28 after bleomycin treatment,5 rats in each group were killed, and the lung were harvested for histopathological studies,immunohistochemical determination of TGF-β1,CTGF,collagenⅠ and collagenⅢ. Results:The degree of alveolitis and fibrosis,the level of TGF-β1,CTGF,collagenⅠ and collagenⅢ decreased significantly in the groups C and groups D, as compared with groups B(P < 0.05). Conclusion:The treatment of Matrine or IFN-γ has some protective effects on pulmonary fibrosis in rats,partly through affecting TGF-β1 and CTGF expression.
