      目的:通过观察视网膜锌离子转运体3(ZnT-3)mRNA表达对低锌饮食的反应,探索低锌是否会引起ZnT-3转录水平的调节?方法:将36只雄性SD大鼠(4周龄)随机分为三大组分别喂养2周(第1组)?4周(第2组)?6周(第3组),每组大鼠又随机分为两个亚组分别给予正常锌含量饮食和低锌饮食,第3组低锌饮食大鼠从第4周开始恢复正常饮食?用逆转录-聚合酶链反应(RT-PCR)法检测ZnT-3以及碳酸酐酶2(CA2)?碳酸酐酶14(CA14)的mRNA相对表达量?结果:大鼠锌摄入低下2周后,与空白对照组相比视网膜CA2和CA14的mRNA水平有所降低,而ZnT-3 mRNA表达量有所增高;锌摄入低下4周后,视网膜ZnT-3?CA2和CA14 mRNA表达量均明显下降;恢复正常锌饮食后2周,实验组视网膜ZnT-3?CA2和CA14的mRNA的表达回升与对照组无差异?结论:饮食锌的变化将导致大鼠视网膜ZnT-3在转录水平上的调节,含锌酶CA2和CA14mRNA表达也同时发生变化?为保持视网膜组织内锌含量的稳定,ZnT-3mRNA表达量随饮食含锌量降低的时间和程度而改变?

      Objective:To examine the transporter 3(ZnT-3) mRNA expression in retina of rats responding to dietary zinc when the dietary zinc is low,and to investigate whether zinc deficient will lead to ZnT-3 expression change at a transcriptional level. Methods:thirty-six male SD rats(4 weeks old) were randomly divided into three groups, the first group(n = 12) was fed for 2 weeks,the second group(n = 12)for 4 weeks and the third(n = 12) for 6 weeks. Then the each group randomly divided into two subgroups,one subgroup(blank control group) given with normal zinc content diet and the other(experimental group) given with low zinc content diet. The third experimental group rats were changed into the diet with normal zinc content from the 4th week to the 6th week. The expression of ZnT-3,carbonic anhydrase 2(CA2) and 14(CA14) were detected by RT-PCR respectively. Results:After the rats took low zinc content diet for 2 weeks,their retina CA2 and CA14 mRNA levels were decreased,and the ZnT-3 mRNA was increased compared with the control; After given the low zinc content diet for 4 weeks,all of the ZnT-3,CA2 and CA14 mRNA levels decreased significantly; Then after being changed back to the normal diet for 2 weeks,the third experimental group rats has ZnT-3,CA2 and CA14 mRNA levels recovery in retina. Conclusion:The result provided evidence that in a zinc deficient animal model,ZnT-3,CA2 and CA14 expression was responsive to zinc at a transcriptional level in retina. To maintain the normal zinc content,the ZnT-3 mRNA expression was regulated by the time and severity of zinc deficiency in rats.
