      目的:检测糖尿病大鼠结肠中Cajal间质细胞(interstitial cells of Cajal,ICC)和血红素氧合酶-2(heme oxygenase 2,HO-2)的改变以及ICC与HO-2的共表达情况,探讨ICC和HO-2在糖尿病结肠动力障碍发生?发展中的可能作用?方法:以免疫组化方法检测大鼠结肠组织中的ICC和HO-2的分布和表达情况;双重免疫荧光标记结肠ICC和HO-2,观察其共表达情况?结果:ICC和HO-2主要分布在大鼠结肠的肌间神经丛,糖尿病大鼠结肠中ICC和HO-2的表达量均较正常大鼠明显减少(P < 0.05);所有大鼠结肠组织ICC阳性细胞和HO-2阳性细胞在同一部位表达,糖尿病大鼠结肠组织中二者共表达的部位较对照组减少?结论:结肠组织ICC和HO-2的分布和表达减少可能是糖尿病结肠运动障碍的原因之一;ICC可能产生一氧化碳并介导一氧化碳在胃肠运动中的作用?

      Objective:To investigate the changes of interstitial cells of Cajal(ICC) and hemeoxygenase-2(HO-2) in the colon of diabetic rats and the immunocolocalization of the ICC and HO-2 in the colon of rats,and discuss the possible mechanism of colon dysfunction associated with diabetes mellitus. Methods:The distribution and expressions of ICC and HO-2 in the proximal and distal colon tissues of rats were detected by immunohistochemistry. Immunolocalization of ICC and HO-2 was detected by confocal laser scanning microscopy. Results:ICC and HO-2 were mainly distributed in the myenteric plexus of the colon tissue,and the expressions of ICC and HO-2 in the proximal and distal colon tissues of diabetic rats were decreased compared with those in the control group(P < 0.05). Double immunofluorescence displayed that c-kit positive cells and HO-2 positive cells were expressed in the identical sites in the colon tissues of rats. Double expressions of ICC and HO-2 in the colon tissues of diabetic rats were decreased compared with those in the control group. Conclusion:Decreased expressions of ICC and HO-2 in the colon tissues might play an important role in the pathogenesis of colon dysfunction associated with diabetes mellitus; ICC may produce CO and intervene its role in the gastrointestinal motility.
