      目的:观察重组人促红细胞生成素(rhEPO)对SD大鼠背部皮瓣的影响,探讨可能的机制?方法:以手术形成大鼠背部任意瓣(2 cm × 9 cm)为模型,应用比色法和硫代巴比妥法观察术后24 h组织中髓过氧化物(MPO)活性及丙二酰二醛(MDA)含量,评价rhEPO对MPO活性和MDA含量的影响;应用大体观察法,测定术后7天皮瓣存活率,评价rhEPO对皮瓣成活率的影响;光镜下观察CD34免疫组化染色切片,评价rhEPO对皮下微血管生成的影响?结果:rhEPO可明显降低术后24 h时MPO活性和MDA含量(P < 0.05),提高术后7天的皮瓣成活率(P < 0.05),增加皮下微血管生成(P < 0.05)?结论:rhEPO是通过减少中性粒细胞浸润,从而提高皮瓣存活率,其具体机制有待进一步研究?
英文摘要:
      Objective:To observe the effects of recombinant human erythropoietin(rhEPO)on skin flap in the SD rats,and discuss the possible mechanism. Methods:A dorsal,caudally based random flap measuring 2 cm × 9 cm was created in all 24 SD rats which were divided into two groups:control group(Group A,n=12)with saline injected through the intraperitoneal route after operation,and treated group(Group B,n=12)with single-dose rhEPO 5 000 IU/kg injected through the intraperitoneal route after operation. At 24 th hour postoperatively,the myeloperoxidase(MPO)activity and the amount of malondialdehyde(MDA) were evaluated on the flap tissue samples of 6 rats chosen randomly. The flap viability and immunohistochemical(CD34)methods which can evaluate the microvascular network were applyed on the rest rats at day 7 after operation. Results:There was significant decrease in MPO and MDA in treated group compared with control group,and a significant increase in the flap viability in treated group was observed(P < 0.05). Conclusion:rhEPO can decrease the MPO activity and amount of MDA in the tissues of random flap at the first 24 h after operation and increase microvessel density and viability at day 7 after operation.
