非小细胞肺癌组织中HIF-1α和BNIP3的表达及相关性研究

    目的:研究缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)和Bcl-2/腺病毒E1B19kDa结合蛋白3(Bcl-2 and adenovirus E1B 19kDa interacting protein 3,BNIP3) 在非小细胞肺癌(non-small cell lung carcinoma,NSCLC)组织中的表达及其相关性-方法:收集45例新鲜NSCLC标本(含癌旁组织)和14例正常对照肺组织,采用半定量RT-PCR和SP免疫组织化学方法,测定HIF-1α和BNIP3的表达水平及分析二者相关性,并与组织类型-分化程度-淋巴结转移-TNM分期-年龄-吸烟等进行多因素分析-结果:肺癌肿瘤组织中HIF-1α-BNIP3的mRNA表达水平显著高于癌旁组织及正常对照肺组织(P < 0.01),并且两者表达呈正相关(r=0.68,P < 0.01);HIF-1α-BNIP3在肿瘤组织中的蛋白阳性表达率分别为53.3%-66.7%,显著高于癌旁组织及正常对照肺组织(P < 0.01),且二者阳性率呈正相关(rs=0.38,P < 0.01)-单因素分析发现HIF-1α与组织类型-TNM分期相关(P < 0.05),而BNIP3在相关临床病理参数分组中无显著差异-结论:HIF-1α与BNIP3在NSCLC中高表达且呈正相关,推测二者的高表达在肺癌的发生发展中可能具有重要作用-

    Abstract:

    Objective:To investigate the expression of hypoxia inducible factor-1α(HIF-1α) and the Bcl-2/adenovirus E1B 19 kDa-interacting protein 3 (BNIP3) in human non-small cell lung carcinoma(NSCLC) and their correlation. Methods:The expression of BNIP3 and HIF-1α in 45 tumor samples,45 para-cancer tissues and 14 control normal lung tissues were examined by reverse transcription-polymerase chain reaction analyses (RT-PCR) and SP immunohistochemical method. Multivariable analysis was used to analyze the correlation of the HIF-1α and BNIP3 expression on histological types,differentiation,TNM clinical staging,lymphatic metastasis,age and cigarette smoking. Results:Compared with the control group,the mRNA levels of HIF-1α and BNIP3 in lung carcinoma tissues were much higher. There was a significant difference between tumors and the control(P < 0.01). BNIP3 mRNA levels were positively correlated with HIF-1α levels(r=0.68,P < 0.01). In lung carcinoma,the positive expression rates of HIF-1α and BNIP3 proteins were 53.3% and 66.7% respectively,which were higher than those in para-cancer tissues and normal lung tissues(P < 0.01). The positive correlation between HIF-1α and BNIP3 expression was highly significant(rs=0.38,P < 0.01). The expression of HIF-1α was correlated with histological types and TNM clinical staging(P < 0.05). No significant correlation was found between BNIP3 expression and patients’ clinicopathologic parameters. Conclusion:The high expression levels of HIF-1α and BNIP3 were closely related in non-small cell lung carcinomas,both of them played important roles in the carcinogenesis and development of lung carcinoma.
