    目的:研究mTOR特异性抑制剂RAD001对胃癌SGC7901细胞增殖的影响,并探讨survivin在其中的作用-方法:常规培养胃癌SGC7901细胞,RAD001单独或联合顺铂干预,MTT法测定RAD001对胃癌SGC7901细胞增殖的影响,流式细胞术检测细胞凋亡率,免疫细胞化学和Western-blot检测药物作用后细胞中survivin蛋白的表达-结果:RAD001单独或联合顺铂干预后,①SGC7901细胞增殖受到抑制,抑制效应呈剂量依赖性并随时间的延长逐渐升高;②高浓度RAD001可以促进SGC7901细胞的凋亡;③SGC7901细胞中survivin蛋白表达下降-结论:RAD001可以通过下调细胞中survivin蛋白的表达抑制胃癌SGC7901细胞增殖,促进其凋亡-

    Abstract:

    Objective: To investigate the role of mTOR specific inhibitor RAD001 in inhibiting gastric cancer cell SGC7901 proliferation and the possible mechanism of survivin in the inhibition. Methods:SGC7901 cells were cultivated, treated with RAD001 alone or in combination with cisplatin. MTT, flow cytometry, Immunocytochemistry and Western-blot array were used to investigate the cell proliferation, apoptosis and the expression of survivin in cells. Results:After treated with RAD001 alone or in combination with cisplatin, the cell proliferation were inhibited in a dose-dependment manner, and the inhibition rates increased with the treatment time extending. High concentration of RAD001 could induce apoptosis of SGC7901 cells. The expression of survivin in SGC7901 cells decreased after treatment. Conclusion:RAD001 could inhibit the cell proliferation of SGC7901 and induce the apoptosis through down-regulating the expression of survivin.
