      目的:探讨TGF-β1和CD105表达在宫颈尖锐湿疣(candyloma acuminatum,CA)和宫颈癌(cervical carcinoma,CC)发生以及演进过程中的作用,评估CD105作为抗血管生成靶分子在治疗宫颈病变中的临床应用价值?方法:用免疫组织化学Envision二步法检测42例CA?50例CC?和20例正常宫颈黏膜中TGF-β1和CD105的表达,采用聚合酶链反应(PCR-荧光法)技术检测宫颈分泌物中人乳头瘤病毒(HPV)DNA?结果:宫颈正常黏膜?CA和CC中HPV DNA检出率分别为20%?100%和92%;TGF-β1表达率分别为30.0%?52.4%和62.0%,差别有统计学意义(P < 0.05)?在宫颈CA和CC中,TGF-β1表达仅与CC分期有显著相关性(P < 0.01)?CD105标记的微血管密度(MVD)在宫颈正常黏膜?CA和CC中存在显著性差异(P < 0.01)?MVD在不同CA病变大小之间存在显著性差异(P < 0.05),在不同CC分级和分期之间存在显著性差异(分别P < 0.01,P < 0.05)?在CA和CC中,TGF-β1表达与MVD存在显著相关性(P < 0.05)?结论:TGF-β1/CD105通路调控的微血管生成在CA和CC发生以及演进过程中发挥重要作用,CD105可以作为较好的抗血管生成治疗的候选靶分子?

      Objective: To evaluate the roles of TGF-β1 and CD105 in the occurrence and development of cervical condyloma acuminateum (CA) and cervical carcinoma (CC),and determine the potential value of CD105 in the antiangiogenic targeted therapy. Methods: The expressions of TGF-β1 and CD105 were investigated by immunohistochemistry Envision method in samples from 42 cases of CA,50 cases of CC,and 20 cases of normal cervical mucosa as controls. The HPV virus was detected by the polymerase chain reaction (PCR-fluorescence method) analysis. Results: HPV DNA was detected in 20% of controls,100% of CA and 92% of CC,respectively (P < 0.01). TGF-β1 was expressed in 30% of controls,52.4% of CA and 62% of CC,respectively (P < 0.05). TGF-β1 expression only positively correlated with the CC clinical stage (P < 0.01). There was a significantly difference of MVD marked by CD105 antibody among cervical CA,CC and controls(P < 0.01). There was a significantly difference between MVD and size of cervical CA (P < 0.05),and histological grade(P < 0.01),and clinic stage of CC (P < 0.05),respectively. A significantly correlation was confirmed between TGF-β1 and MVD in both CA and CC(P < 0.05). Conclusion: These data provide evidences that TGF-β1/CD105 system plays an important role in abnormal micro-angiogenesis in the occurrence and progress of cervical CA and CC. CD105 could be taken as a potential antiangiogenic target in the treatment of cervical CA and CC.
