    目的:探讨H2S对巨噬细胞COX-2表达及泡沫化的影响-方法:建立氧化低密度脂蛋白(Ox-LDL)诱导RAW264.7细胞泡沫化模型,同时给予不同浓度的H2S-应用油红O染色观察泡沫细胞的形成;Real-time qPCR检测COX-2 mRNA的表达;Western blot检测COX-2蛋白的表达-结果:Ox-LDL能够明显增加细胞内脂质的沉积,同时伴有COX-2的产生-不同浓度的H2S可以明显减少由Ox-LDL引起的COX-2的表达,同时也减少了细胞内脂质的沉积,以250 μmol/L最为明显,差异具有统计学意义(P < 0.05)-结论:H2S可以显著减少巨噬细胞COX-2的表达及泡沫化-

    Abstract:

    Objective:To explore effects of H2S on expression of COX-2 on macrophages(RAW264.7) and formation of macrophage-derived foam cells. Methods:RAW264.7 foam cell model induced by low-density lipoprotein (Ox-LDL) is established,then added with different concentrations of H2S. Detection of foam cell formation is conventionally practiced by Oil Red O staining. The expressions of COX-2 mRNA and protein are detected by real-time qPCR and Western blot respectively. Results:A significant increase of the production of COX-2 and the deposition of intracellular lipid was observed. Different concentrations of H2S can considerably reduce the expression of COX-2 and the deposition of intracellular lipid induced by the Ox-LDL,especially the concentration of 250 μmol/L and the difference is statistically significant(P < 0.05). Conclution:H2S can significantly reduce the expression of COX-2 in macrophages(RAW264.7 )and inhibit the formation of foam cells .
