      目的:研究环境内分泌干扰物双酚A(BPA)对SD大鼠血-脑屏障成分表达及星形胶质细胞活力的影响并探讨其损伤机制?方法:通过MTT检测细胞活力,高效液相色谱法(HPLC)检测谷氨酸含量的变化,实时荧光定量PCR检测星形胶质细胞中Claudin-3?Claudin-4?Claudin-5?JAM-1?JAM-2?ZO-1?E-cadherin基因的表达?结果:10-7 mg/ml 的BPA与星型胶质细胞孵育24或48 h后即可引起细胞活力的下降,具有统计学差异?在该浓度的暴露下,血-脑屏障紧密连接相关蛋白Claudin-3?Claudin-4?JAM-1?E-cadherin 蛋白的mRNA水平下调,而Claudin-5?JAM-2?ZO-1蛋白基因表达上调且具有统计学意义;HPLC结果显示,谷氨酸的浓度上调?结论:环境内分泌干扰物BPA对血-脑屏障的完整性具有潜在的损伤作用?

      Objective:To investigate the major effects of environmental endocrine disruptors (EDCs) BPA on SD rat blood-brain barrier component expression,cell viability of astrocytes and explore the mechanism. Methods:The MTT assay was performed to evaluate the astrocyte viability,HPLC was performed to detect glutamate levels,and by real-time quantitative PCR the mRNA levels of Claudin-3,Claudin-4,Claudin-5,JAM-1,JAM-2,ZO-1 and E-cadherin were detected. Results:After astrocytes were exposed to BPA for 24 or 48 hours,the cell viability significantly decreased. The realtime-PCR data showed that mRNA levels of blood-brain barrier tight junctional protein,Claudin-3,Claudin-4,JAM-1,E-cadherin,were significantly down-regulated. Meanwhile,Claudin-5,JAM-2 and ZO-1 mRNA expressions were significantly up-regulated. Furthermore,HPLC showed that the concentration of glutamate was increased. Conclusion:Taken together,EDCs BPA demonstrated the potential toxic effects on the blood-brain barrier integrity.
