    摘要:

    目的:探讨褪黑素(melatonin,MT)对两种横纹肌肉瘤细胞系RD、RH30增殖、迁移、侵袭的影响及其作用机制。方法:MT处理RD与RH30细胞系,分别通过MTT、EdU、Transwell等方法检测两组细胞的增殖、迁移与侵袭的情况。Western blot检测MT作用前后细胞Sonic Hedgehog(Shh)信号通路下游转录因子Gli1表达水平的变化。在两组细胞中分别抑制和过表达Gli1后,检测细胞增殖、迁移与侵袭的情况改变。结果:MT能有效抑制RH30细胞的增殖、迁移与侵袭,但对RD细胞并无明显作用,并且MT受体抑制剂Luzindole不改变MT对RH30增殖的抑制作用。MT处理后RH30细胞中Gli1的表达呈下降趋势,且改变Gli1表达后细胞增殖、迁移、侵袭都发生了改变。结论:MT能够显著抑制RH30细胞的增殖、迁移和侵袭能力,其作用机制与MT能够抑制RH30细胞中Gli1的表达水平有关。

    Abstract:

    Objective:To investigate the effect of melatonin on proliferation,migration and invasion of two rhabdomyosarcoma cell lines RD and RH30 and its mechanism. Methods:The proliferation,migration and invasion of both groups were detected by MTT,EdU and Transwell,respectively. Western blot was used to detect the expression of Gli1 in the downstream transcription factor of sonic hedgehog (Shh) before and after MT treatment. After inhibition and over-expression of Gli1 in the two groups,cell proliferation,migration and invasion were detected. Results:MT effectively inhibited the proliferation,migration and invasion of RH30 cells,but had no obvious effect on RD cells,and luzindole,the MT receptor inhibitor,did not change the inhibitory effect of MT on RH30 proliferation. The expression of Gli1 in RH30 cells decreased after MT treatment,and the cell proliferation,migration and invasion changed after Gli1 expression was changed. Conclusion:MT can significantly inhibit the proliferation,migration and invasion of RH30 cells,and its mechanism is related to the inhibition of Gli1 expression in RH30 cells by MT.
