      目的:探究甘氨酸对华法林诱导的瓣膜间质细胞(aortic valve interstitial cells,AVICs)钙化是否有保护作用。方法:提取猪AVICs,通过钙浓度测定、碱性磷酸酶(alkaline phosphatase,ALP)活性测定、茜素红染色等方法检测不同浓度甘氨酸对钙化的作用;real?time PCR探究甘氨酸对成骨分化的作用;Western blot、流式细胞学及TUNEL(TdT?mediated dUTP Nick?End Labeling)染色研究甘氨酸对华法林引起的AVICs凋亡的作用。结果:甘氨酸抑制华法林引起的AVICs钙化,抑制mRNA水平成骨分化标志物的表达,抑制Smad1、5/RUNX2通路,并且抑制华法林引起的AVICs凋亡。结论:甘氨酸可能抑制Smad1、5/RUNX2通路,并且抑制钙化早期凋亡,因此对华法林引起的AVICs钙化具有保护作用。
英文摘要:
      Objective:To investigate if glycine has the protection effect on warfarin induced aortic valve interstitial cells(AVICs)calcification. Methods:Porcine AVICs were isolated,calcium quantification,alkaline phosphatase activation and Alizarin red staining were performed to determine the effect of different concentrations of glycine on calcification,and the mRNA expression of osteogenic markers were determined by RT?PCR. Western blot,Flow cytometry and TUNEL assay were performed to investigate the anti?apoptosis effect on warfarin induced AVICs calcification. Results:Glycine attenuated warfarin induced AVICs calcification,reduced the mRNA expression of osteogenic markers,inhibited the Smad1,5/RUNX2 pathway and decreased the apoptosis of warfarin induced calcification. Conclusion:Glycine inhibits Smad1、5/RUNX2 pathway and decreases the early stage of apoptosis to attenuate warfarin induced AVICs calcification.
