                环状RNA hsa_circ_0001479在胃癌中的表达及功能研究

               [摘   要] 目的:探讨hsa_circ_0001479在胃癌(gastric cancer,GC)组织中的表达情况及其对胃癌细胞生物学行为的影响。方
                法:建立实时荧光定量PCR(RT⁃qPCR)检测hsa_circ_0001479表达量的方法并进行方法学性能评价;通过RT⁃qPCR检测76对
                胃癌组织和配对癌旁组织中 hsa_circ_0001479 的表达水平并分析其表达量与临床病理参数的关系;构建针对 hsa_ circ_
                0001479的小干扰RNA(small interfering RNA,siRNA)和过表达载体,转染至胃癌细胞株,通过CCK⁃8实验、克隆形成实验检测
                细胞增殖能力,流式细胞术检测细胞周期分布,划痕实验检测细胞横向迁移能力,Transwell实验检测细胞侵袭和迁移能力。结
                果:RT⁃qPCR 检测 hsa_circ_0001479 表达量的方法具有良好的精密度、正确度和线性;hsa_circ_0001479 在胃癌组织中表达增
                高且表达水平与肿瘤大小、浸润深度、淋巴结转移、TNM 分期和 CA19⁃9 水平有关;细胞功能实验显示,在胃癌细胞株中干扰
                hsa_circ_0001479的表达可抑制胃癌细胞的增殖、侵袭和迁移,过表达hsa_circ_0001479则呈现相反的结果。结论:胃癌组织中
                高表达的hsa_circ_0001479具有促进胃癌细胞增殖、侵袭和迁移的功能。

                Expression and function of hsa_circ_0001479 in gastric cancer

               [Abstract] Objective:The current study aims to explore the expression of hsa_circ_0001479 in gastric cancer(GC)tissues and its
                effect on biological behavior of GC cells. Methods:The method for detecting the expression of hsa_circ_0001479 with real ⁃ time
                fluorescence quantitative polymerase chain reaction(RT⁃qPCR)was established and evaluated. Expression level of hsa_circ_0001479
                in 76 pairs of GC and para ⁃ cancerous tissues was measured by RT ⁃ qPCR and relationship between the expression level and
                clinicopathological factors was analyzed. Small interfering RNA(siRNA)and overexpression vector of hsa_circ_0001479 were
                constructed and transfected into GC cell lines. Cell proliferation was detected by CCK⁃8 assay and clone formation assay. Cell cycle
                distribution was detected by flow cytometry. Cell migration was detected by wound healing assay and Transwell assay. Cell invasion was
                detected by transwell assay. Results:Using RT ⁃ qPCR to detect the expression of hsa_circ_0001479 possessed good precision,
                accuracy and linear range. Hsa_circ_0001479 was upregulated in GC tissues and the expression level was related to tumor size,depth
                of invasion,lymph node metastasis,TNM stage and CA19⁃9. Silencing of hsa_circ_0001479 inhibited the proliferation,invasion and
                migration of GC cells while overexpression of hsa_circ_0001479 showed the opposite results. Conclusion:The carrent study found that
                hsa_circ_0001479 was upregulated in GC tissues and could promote GC cell proliferation,invasion and migration.
